Ford f250 on 37s.
That half inch up front won't make a difference whatsoever. The 2020 F250 uses a shorter rear block, looks to be 2" vs. the 17-19 3.25". That difference again, has no affect on 37's but might give a saggy rear look with 2.5" up front. You said adding rear blocks, if needed, isn't a problem.

I had a 3" rough country lift with 37's on 20" factory wheels. The tires did not hit the wheel wells but they did rub the frame pretty bad at full turn. The tires were 37x12.50 nitto trail grappler. You can do all the lift you want but the tires are going to rub the frame with the factory wheels because of the off set.

These trucks will run a 35 in stock form. I have 37s on mine right now. It has 2.5" ready lift all the way around. My 37s are on 20x10 Fuel Mavericks. Has anyone successfully placed 37" bfg km2's on a f250 with a 4" suspension lift with little to no modifications to the truck. I can squeeze 35's on my stock height truck, so i don't see why I couldn't on a 4' lift.

I think 4.56 would be too tall, I’d recommend 4.30, good balance for towing and mpgs. I would go at least 4.88's if you tow anything remotely heavy.

I've got the 2.5" Readylift and 37x12.5's on the stock wheels. They do rub the radius arms but its not bad enough to worry about. Go with 295/65-20. FYI - Ford is putting a 35" spare under the new Tremor package trucks. So the weight difference you're talking is maybe 5-10 lbs. Carli arms really are more about bling than function. They are notched to give 1/2" additional clearance at full steering lock. This is completely a non-issue with greater offset wheels and 37s.These trucks will run a 35 in stock form.
